I have a 2000 V6 Dakota. The check engine comes on sometimes and when we checked the codes it comes up P0303 and P0304. What does this mean?

P0303 Cylinder 3 Misfire Detected

P0304 Cylinder 4 Misfire Detected

maybe bad spark plugs, plug wires, fuel injectors? How many miles are on it and how long ago did you do a tune-up?

Dan ol' buddy, you will not get a misfire signal if the fuel injectors are bad. It only detects a spark cycle and sends a misfire code if there is something wrong with the spark discharge. There is a separate code if the injectors mis-spurt.

Doh? A cylinde misfire condition will be detected if any of the necessary components for proper cylinder operation are not adequate ie not enough fuel, low compression and no spark.

A spark discharge malfunction is not the only reason for a cylinder misfire.

Yes, there are specific codes for injector issues but a failing injector might not throw a code just as a failing o2 sensor (just an example as there have been alot of discussion on o2 sensors lately) won't throw a code. It's within spec (proper electrical operation) but enough to be off to cause other systems/components to malfunction.

refer to P0300 - random/multiple cylinder misfire.

Notice it was 3rd in the list behind plugs and wires. It's more than likely not the cause but a possibility.
