I have an 02 dakota 4x4 4.7L and the rear end(9.25 limited slip) is making noise when I turn from a stop. The noise sounds like a loud clicking and I can feel vibrations in the truck. Any help would be greatly appreciated

When was the fluid last serviced (how many miles since then) and was the additive put into the diff?

Without hearing the noise: hard to say.

I changed the diff fluid about 20,00 miles ago and I put the friction modifier in. My truck has 49,900 miles on it. Should I try changing the fluid again?

I had a noise like that from my jeep's rear end. It turned out to be one of the side ring gears to be broken in half. Made a clacking sound as it wobbled. It got worse and eventually jammed and locked the gears essentially stopping wheel travel and I skidded to a stop. If I backed up a bit they would unlock, but eventually it had to be dragged onto a tilt bed and hauled to the dealer who wanted $1,500. I took it to an independant drive train shop and paid $750. So when my 99' Dakota started making noise like it I knew what it was and it turned out to be broken gears in it too. Hopefully yours will be something cheaper to fix, but don't rule out broken gear set.
